  Affinity Measurement by Biolayer Interferometry

CPTC-GPC6-1 Affinity and Kinetics (Bio-Layer Interferometry)

Result: No Binding

Affinity and binding kinetics of CPTC-GPC6-1 antibody and full-length GPC6 recombinant protein were measured using biolayer interferometry. GPC6 full length recombinant protein was amine coupled onto AR2G biosensors. CPTC-GPC6-1 antibody was used as analyte and was titrated at 1024 nM, 256 nM, 64 nM, 16 nM, 4 nM, 1 nM, and 0.25 nM. Buffer only and biosensors having no recombinant protein were used as references for background subtraction. All binding data were analyzed globally.

The antibody was able to pull down the target in cell lysates HeLa, HepG2, SF-539, SNB-75 and SW620, but not in HL-60 cell lysate (top panel). IP results were compared with cell lysates direct analysis in SW, using CPTC-GPC-1 as detection antibody (bottom panel). Click image to enlarge

Background

NCI Identification Number:

00540

Antigen Name:

Glypican 6

CPTC Name:

CPTC-GPC6

Aliases:

Glypican 6; Glypican Proteoglycan 6; Glypican-6; OMIMD1

Function:

The glypicans comprise a family of glycosylphosphatidylinositol-anchored heparan sulfate proteoglycans, and they have been implicated in the control of cell growth and cell division. The glypican encoded by this gene is a putative cell surface coreceptor for growth factors, extracellular matrix proteins, proteases and anti-proteases. Mutations in this gene are associated with omodysplasia 1.GPC6 (Glypican 6) is a Protein Coding gene. Diseases associated with GPC6 include Omodysplasia 1 and Omodysplasia. Among its related pathways are Apoptotic Pathways in Synovial Fibroblasts and GPCR Pathway. Gene Ontology (GO) annotations related to this gene include heparan sulfate proteoglycan binding. An important paralog of this gene is GPC4.Cell surface proteoglycan that bears heparan sulfate. Putative cell surface coreceptor for growth factors, extracellular matrix proteins, proteases and anti-proteases (By similarity). Enhances migration and invasion of cancer cells through WNT5A signaling.
