The affinity and binding kinetics of CPTC-STAT3-2 antibody and human signal transducer and activator of transcription 3 (acute-phase response factor) (STAT3) recombinant protein, transcript variant 1, were measured using biolayer interferometry. Recombinant protein was covalently immobilized onto AR2G biosensors using standard amine coupling. CPTC-STAT3-2 antibody at 1024 nM, 64 nM, 16 nM, 4 nM, 1 nM, and 0.25 nM, was used as analyte. Buffer only and biosensors immobilized without recombinant protein were used as references for background subtraction. All data was analyzed globally using a 1:2 bivalent model.

Binding data were double-referenced and analyzed globally using a bivalent model.

Flow cytometric analysis of STAT3 expression in SN12C and COLO205 cells using CPTC-STAT3-2 rabbit antibody. SN12C cells were permeabilized, fixed and then stained with CPTC-STAT3-2 antibody (solid green) or concentration-matched rabbit isotype control antibody (dashed green). COLO205 cells were permeabilized, fixed and then stained with CPTC-STAT3-2 (solid blue) or concentration-matched rabbit isotype control antibody (dashed blue) and then fixed. A BV421 conjugated goat anti-rabbit IgG (H+L) was used as a secondary antibody. All data were analyzed using FlowJo. CPTC-STAT3-2 antibody can detect expression of STAT3 in both SN12C and COLO205 cells.

Immunoprecipitation using CPTC-STAT3-2 as capture antibody to probe the lysates of SNC12, IGROv1 and MCF7. Eluates were tested in Simple Western, using CPTC-STA3-2 as primary antibody (Panel A). The antibosy was not able to pull down the target protein, as also evident in the comparison between input material (blue line) and eluate (green line) profiles in panels B, C and D, respectivelly for SNC12C, IGROV1 and MCF7.

The protein encoded by this gene is a member of the STAT protein family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is activated through phosphorylation in response to various cytokines and growth factors including IFNs, EGF, IL5, IL6, HGF, LIF and BMP2. This protein mediates the expression of a variety of genes in response to cell stimuli, and thus plays a key role in many cellular processes such as cell growth and apoptosis. The small GTPase Rac1 has been shown to bind and regulate the activity of this protein. PIAS3 protein is a specific inhibitor of this protein. Mutations in this gene are associated with infantile-onset multisystem autoimmune disease and hyper-immunoglobulin E syndrome.
