Immuno-MRM chromatogram of CPTC-CD70-1 antibody (see CPTAC assay portal for details: https://assays.cancer.gov/CPTAC-5959)
Data provided by the Paulovich Lab, Fred Hutch (https://research.fredhutch.org/paulovich/en.html). Data shown were obtained from FFPE tumor tissue lysate pool.
Result: Positive
Protein Array in which CPTC-CD70-1 is screened against the NCI60 cell line panel for expression. Data is normalized to a mean signal of 1.0 and standard deviation of 0.5. Color conveys over-expression level (green), basal level (blue), under-expression level (red).
Result: Negative
Western blot using CPTC-CD70-1 as primary antibody against human CD70 molecule (CD70) recombinant protein (lane 2). Expected molecular weight - 20.9 kDa. Molecular weight standards are also included (lane 1).
Result: Positive
Western blot using CPTC-CD70-1 as primary antibody against A549 (lane 2), HeLa (lane 3), Jurkat (lane 4), MCF7 (lane 5), H226 (lane 6), and PBMC (lane 7) whole cell lysates. Expected molecular weight - 21.1 kDa and 23.4 kDa. Molecular weight standards are also included (lane 1). A549, HeLa and MCF7 are positive. All other cell lines are negative. Target protein is subject to glycosylation which can affect the migration in electrophoresis. This can make the target appear as a higher molecular weight protein.

Function:
The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for TNFRSF27/CD27. It is a surface antigen on activated, but not on resting, T and B lymphocytes. It induces proliferation of costimulated T cells, enhances the generation of cytolytic T cells, and contributes to T cell activation. This cytokine is also reported to play a role in regulating B-cell activation, cytotoxic function of natural killer cells, and immunoglobulin sythesis. CD70 (CD70 Molecule) is a Protein Coding gene. Diseases associated with CD70 include Lymphoproliferative Syndrome 3 and Acute Myocarditis. Among its related pathways are PEDF Induced Signaling and TNF Superfamily - Human Ligand-Receptor Interactions and their Associated Functions. Gene Ontology (GO) annotations related to this gene include signaling receptor binding and protease binding.Cytokine which is the ligand for CD27. The CD70-CD27 pathway plays an important role in the generation and maintenance of T cell immunity, in particular during antiviral responses. Upon CD27 binding, induces the proliferation of costimulated T-cells and enhances the generation of cytolytic T-cells.
