iMRM data obtaiend form the incubation of the cell lysate H3255 digest with clone CPTC-ERRFI1-1. Data were acquired on the nano-chip-LC using a 1260 Infinity Series HPLC-Chip cube interface (Agilent, Palo Alto, CA) coupled to a 6495-triple quadrupole mass spectrometer (Agilent). A large capacity chip system (G4240-62010) consisting of a 160 nl enrichment column and a 150 mm*75 um analytical column (Zorbax 300SB-C18, 5 um, 30 A pore size) was used. The clone is able to capture the endogenous target peptide VSSTHYpYLLPERPPYLDK (CPTC-ERRF1 Peptide 1).
Result: Negative
Immunofluorescence staining of human cell line A549 with CPTC-ERRFI1-1 Ab shows no localization of ERRFI1 protein.
Result: Negative
This antibody is not suitable for use in a Reverse Phase Protein Array format as described in SOP M-105.
Result: Positive
Western Blot usnig CPTC-ERRFI1-1 against the ERRFI1 Overexpressed Cell Lysate. Expected MW is about 50 KDa
Result: Positive
Western Blot using CPTC-ERFFI1-1 as primary antibody against cell lysates OVCAR3, HCT116, PC-3, MBA-MD-231 and A549 (lane 2) with expected MW of 50 KDa. Positive for A549 lysates, inconclusive/negative results for the other cell lysates. Molecular weight standards are also included (lane 1). ECL detection.
Result: Negative
Single cell western blot using CPTC-ERRFI1-1 as a primary antibody against cell lysates. ERRFI1 protein was not detected and therefore no expression graphs were generated.

Antigen Name:
ERBB Receptor Feedback Inhibitor 1 Peptide 1
CPTC Name:
CPTC-ERRFI1 Peptide 1
Aliases:
ERBB Receptor Feedback Inhibitor 1; MIG-6; Mitogen-Inducible Gene 6 Protein; GENE-33; MIG6; RALT; Receptor-Associated Late Transducer; ERRFI1
Function:
ERRFI1 is a cytoplasmic protein whose expression is upregulated with cell growth (Wick et al., 1995 [PubMed 7641805]). It shares significant homology with the protein product of rat gene-33, which is induced during cell stress and mediates cell signaling (Makkinje et al., 2000 [PubMed 10749885]; Fiorentino et al., 2000 [PubMed 11003669]).ERRFI1 (ERBB Receptor Feedback Inhibitor 1) is a Protein Coding gene. Diseases associated with ERRFI1 include Endometrial Disease and Progesterone Resistance. Among its related pathways are EGF/EGFR Signaling Pathway and Tyrosine Kinases / Adaptors. Gene Ontology (GO) annotations related to this gene include protein kinase binding and kinase binding.Negative regulator of EGFR signaling in skin morphogenesis. Acts as a negative regulator for several EGFR family members, including ERBB2, ERBB3 and ERBB4. Inhibits EGFR catalytic activity by interfering with its dimerization. Inhibits autophosphorylation of EGFR, ERBB2 and ERBB4. Important for normal keratinocyte proliferation and differentiation. Plays a role in modulating the response to steroid hormones in the uterus. Required for normal response to progesterone in the uterus and for fertility. Mediates epithelial estrogen responses in the uterus by regulating ESR1 levels and activation. Important for regulation of endometrium cell proliferation. Important for normal prenatal and perinatal lung development (By similarity).
