Affinity and binding kinetics of CPTC-NUDT16L1-1 and BSA-conjugated peptide, CLVEALAAATEK, were measured using surface plasmon resonance. BSA-conjugated peptide was amine coupled onto a Series S CM5 biosensor chip. CPTC-NUDT16L1-1 rabbit antibody was used as analyte and was titrated at 1024 nM, 256 nM, 64 nM, 16 nM, 4 nM, 1 nM, 0.25 nM, and 0.0625 nM. All binding data was double referenced and analyzed globally using a 1:1 fitting model.
Result: Negative

Function:
NUDT16L1 (Nudix Hydrolase 16 Like 1) is a Protein Coding gene. Diseases associated with NUDT16L1 include Encephalopathy, Ethylmalonic and Spastic Hemiplegia. Among its related pathways are Proteoglycans in cancer and Syndecan-4-mediated signaling events. Gene Ontology (GO) annotations related to this gene include hydrolase activity. An important paralog of this gene is NUDT16.Key regulator of TP53BP1 required to stabilize TP53BP1 and regulate its recruitment to chromatin (PubMed:28241136). In absence of DNA damage, interacts with the tandem Tudor-like domain of TP53BP1, masking the region that binds histone H4 dimethylated at 'Lys-20' (H4K20me2), thereby preventing TP53BP1 recruitment to chromatin and maintaining TP53BP1 localization to the nucleus (PubMed:28241136). Following DNA damage, ATM-induced phosphorylation of TP53BP1 and subsequent recruitment of RIF1 leads to dissociate NUDT16L1/TIRR from TP53BP1, unmasking the tandem Tudor-like domain and allowing recruitment of TP53BP1 to DNA double strand breaks (DSBs) (PubMed:28241136). Binds U8 snoRNA (PubMed:18820299).
